Viagra’s benefits continue – may protect heart from high blood pressure
Sildenafil, the key ingredient in Viagra, may shield the heart from high blood pressure damage according to a new study of mouse hearts from Johns Hopkins University.
The researchers say sildenafil appears to affect RGS2 (regulator of G-protein signalling 2), a protein essential to reactions that protect the heart’s blood pumping function from escalating into heart failure, according to MedlinePlus.
The findings were published in The Journal of Clinical Investigation.
"Sildenafil clearly prolongs the protective effects of RGS2 in mouse hearts," says senior investigator Dr. David Kass.
The team induced high blood pressure in the mice for a week, and found hearts modified to lack RGS2 expanded in weight by 90%, with almost half of the animals dying of heart failure. Mice with RGS2 delayed hypertrophy, or dangerous muscle expansion; growing by just 30% - and none of those mice died.
Further testing showed hypertensive mice given sildenafil exhibited less hypertrophy, enhanced buffering, and stronger heart muscle contraction and relaxation. The mice also had around 10 times lower stress-related enzyme activity. Mice lacking RGS2 were unaffected by sildenafil.
"The evidence is piling up that unbridled Gq signaling is driving a central biological chain reaction in heart failure, and that by extending the protective effects of RGS2 or by developing a test for its presence, researchers can develop new therapies or improve existing ones, including ACE inhibitors and possibly sildenafil, for people with heart failure who will benefit most," Kass said.
ACE inhibitor and ARB inhibitor drugs are currently used to block Gq signalling, and are the most common treatment for heart failure. Cardiovascular disease (heart, stroke and blood vessel disease) is the leading cause of death in New Zealand accounting for 40% of deaths annually (approximately 11,300).

The history of the development of Viagra is interesting and should put this article in context.
Viagra was originally developed to treat systemic hypertension (high blood pressure), and to treat angina pectoris ('heart-related chest pain'), however was found to have little effect on these in the initial (phase 1) human trials. The story goes that the investigators struggled to get male participants to return their medications at the conclusion of the trial because of the erectile effects which led to the development and subsequent patenting and licensing for treatment of erectile dysfunction. The drug has been thought to have little potential as a treatment for hypertension due to the results of these initial trials, but in practical terms because the revenue potential from the erectile dysfunction application far outperformed the hypertension application, at a higher wholesale price-per-pill- meaning that the use of viagra for treating hypertension was cost-inefficient.
Since it was originally licensed in NZ, viagra has been used for a type of hypertension called pulmonary hypertension and is know to be one of the few effective therapies.
Thus, the study cited in today's NBR article is irrelevant in the wider context of the history of Viagra.
Not really interested in the heart scenario.
What I want to know is when is the price going to come down, or when will the generic equivilent be available at a reasonable price.
I think that the drug company has made its profit, considering that the erectile dysfunction aspect was a side effect of another proposed developement. In other words a lucky discovery.
I do not think we as men should have to suffer the costs of a dysfunction that is not our fault to profit a company that accidentaly discovered a treatment that can aleviate it.
